Standard Hybridoma Development Program Abeome's DiSH technology is faster than conventional technologies and has been shown to produce more positive, stable hybridoma clones. The Company's Standard Hybridoma Development Program (SHDP) is priced at $9,000.00 per antigen. For pricing of Optional Services connected with SDHP, click here. Preparation of antigen Client provides approximately 1.5 mg of antigen required for the project. Abeome can provide selected antigen preparation services described in Optional Services. Immunizations and boosting Abeome will immunize three mice per antigen in accordance with ALAC and IACUC guidelines. Test bleeds (by ELISA) are done on each animal prior to project initiation; results are available on request. Immunization will be done using TiterMax adjuvant; alternative adjuvants can be used on request. Boosts are given at approximately 28-day intervals. Client is shown all test bleed results and asked to decide whether to boost or harvest. Harvest and fusion Spleens will be harvested, processed, and fused with Abeome's proprietary Ig and Ig -expressing myeloma. Client may choose between the SP2ab or NSab myelomas, derived, respectively from the SP2/0 and NS0 myelomas. (NOTE: Should the Client wish to provide either a pre-immunized mouse, their spleens or spleen cells previously prepped, Abeome will adjust program fees accordingly.) Clone selection and culturing Hybridomas are screened using fluorescent activated cell sorting instrumentation, with reactive hybridomas deposited as single cells into 96-well culture plates. Colonies arising from sorted cells are then screened by ELISA for reactivity with the Client's antigen. Positive colonies are screened for stability, isotyping by ELISA, and level of surface expression. Screening results are shared with the Client who may decide which clones (up to six clones for a three-mouse Program) they wish to pursue. Client-selected clones are grown in culture such that six vials of 2-7×106 cells/vial are frozen for delivery. Culture supernatants (up to 50 ml total) are also delivered to the Client. Storage of hybridomas Abeome will store one vial of each selected clone for up to one year for no charge to the Client. Return to top
